  While bacteria and eukaryotes show distinct mechanisms of DNA damage response (DDR) regulation, it remains elusive what strategy has adopted to deal with DNA damage repair in Archaea, the third domain of life. Recently, our investigation on the DDR regulation in Sulfolobus islandicus by RNA-seq analysis has revealed an Orc1-2-centered DDR regulatory network in this organism. The master DDR regulator can either activate or repress the expression of a large number of DDR genes as for eukaryotic DDR regulators, in contrast to the derepression strategy employed in bacteria. Promoter switch experiment showed that a high level of Orc1-2 is essential, but not sufficient, for the initiation of DDR regulation, suggesting that the regulator is subjected to activation probably by posttranslational modifications (PTMs). Further experiments show that: (a) Orc1-2 interacts with a conserved hexanucleotide motif in the promoter regions of DDR genes and regulates their expression, and (b) protein-protein interaction is probably regulated by DNA damage- activated PTMs, in analogy to regulatory logic in the eukaryotic DDR system. In conclusion, the DDR mechanisms discovered in S. islandicus show a striking similarity to those in eukaryotes, which could represent an archetypal DDR system for the archaeal and eukaryotic lineage of organisms.
